Newborn Calf Serum Testing

According to the provisions of the ChP, newborn calf serum is obtained by separating the blood from newborn calves that have not eaten within 14 hours of birth and then sterilizing and filtering. No other substances may be added during the production process. Newborn calf serum must undergo a series of inspections before use to ensure that its quality meets the following standards:

1. pH value: should be 00~8.50.
2. Protein content: measured by biuret method or other appropriate methods, should be 35~50g/L.
3. Hemoglobin: measured by spectrophotometry or other appropriate methods, should not be higher than 200mg/L.
4. Osmolality: should be 250~330mOsmol/kg.
5. Bacterial endotoxin test: should not more than 10EU/ml.
6. Support cell proliferation test: use passage cells to ensure good cell growth.
7. Sterility test: Check According to the method, should meet the requirement.
8. Mycoplasma test: According to the method, should meet the requirement.
9. Escherichia coli phage: According to plaque method and proliferation method, no bacteriophage contamination.
10. Virus test: According to Cell culture method and Fluorescent antibody method, and the result should be negative.
